No, that should work out of the box. The default T2 initrd contains the USB drivers, even firewire and SBP2, as well as includes a 15 seconds "wait for rootdevice" loop. with root=/dev/sd<the usb number> it works for me (e.g. also for the livecd case where it boots from USB CD-ROMs. Do you run into a specific problem?
