Have you confirmed that local Administrator has access to that key in the first place? Or was it local admin that you created the key manually with?

Maybe your Active Directory is overwriting  the local permissions. We had this problem here.

There are some programs that could prohibit registry edits unless specifically
allowed - like the Cisco Security Agent.  Anything like that installed or
configured?
